Antibody detects endogenous levels of Tau only when phosphorylated at Serine 235.


Target Information

Tau is a neuronal microtubule-associated protein predominantly found on axons. It promotes tubulin polymerization and stabilizes microtubules.
The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, acting as a linker between both.
Axonal polarity is determined by TAU/MAPT localization. Short isoforms allow cytoskeletal plasticity, while longer isoforms contribute to stabilization.
In its hyper-phosphorylated form, Tau is the major component of paired helical filaments (PHF), which form neurofibrillary lesions in Alzheimer’s disease (AD) brains.
Hyper-phosphorylation impairs microtubule binding, destabilizing microtubules and leading to neuronal degeneration.
Kinases such as GSK-3β, PKA, cdk5, and casein kinase II phosphorylate Tau.
Hyper-phosphorylated Tau is found in neurofibrillary lesions of various CNS disorders including Pick’s disease, frontotemporal dementia, cortico-basal degeneration, and progressive supranuclear palsy.
