Antibody detects endogenous levels of Androgen Receptor only when phosphorylated at Ser651, which site historically referenced as Ser650.

Target Information

The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. It is a >90 kDa protein with three major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain. AR is a ligand-activated transcription factor that binds testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, AR dissociates from accessory proteins, translocates into the nucleus, dimerizes, and activates transcription of androgen-responsive genes.

The AR signaling pathway is essential for development and function of male reproductive organs (e.g., prostate, epididymis) and also functions in nonreproductive tissues such as muscle, hair follicles, and brain. AR is a phosphoprotein that regulates MAP kinase activity; inhibition of the MEK1/2 pathway correlates with changes in AR phosphorylation. Dysregulation of AR signaling is associated with prostate cancer, Kennedy’s disease, male infertility, and complete androgen insensitivity syndrome (CAIS).
