Product Specific Information

The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha).
CD127 complexes with CD132 (common gamma chain) to form the IL-7 receptor (IL-7R).
It is expressed on immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation.

IL-7 binding triggers signal transduction via tyrosine kinase pathways including Jak/STAT.
IL-7 is essential for lymphocyte development and T-cell homeostasis.
CD127 expression is down-regulated on CD4+CD25+ regulatory T cells (Tregs); thus, low CD127 expression correlates with Foxp3+ suppressive Tregs.

Binding of eBioRDR5 antibody to PBMCs is blocked by recombinant human IL-7 (Product #14-1079-80).
Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Tested: Pre-titrated for flow cytometry at 5 µL (0.6 µg)/test (100 µL final volume, 10⁵–10⁸ cells/test)

NovaFluor dyes are not compatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

CellBlox Plus Blocking Buffer (Product #C001T06F01)
Use 5 µL per sample for optimal staining. Add to antibody cocktails prior to mixing with cells.

Target Information

CD127 (Interleukin-7 receptor alpha, IL7Rα) is a glycoprotein regulating lymphopoiesis.
The active receptor is a heterodimer of alpha/gamma chains.
CD127 promotes proliferation of precursor B cells, thymocytes, T cell progenitors, and mature CD4+/CD8+ T cells.
Dysfunction of CD127 is associated with severe combined immunodeficiency (SCID).
