Product Specific Information

The monoclonal antibody HP-F1 recognizes CD85j, also known as ILT2, LILRB1, and LIR1.
CD85j is a member of the ILT/LIR/MIR family, a single transmembrane glycoprotein with a long cytoplasmic domain containing four ITIMs that signal through SHP-1 interactions.

• Expression: Found on myeloid cells (monocytes, dendritic cells) and some lymphoid cells (NK, T, B cell subsets).
• Clinical relevance: Correlated with leukemias such as ALL and CLL.
• Functional role: Expression on CD8+ cells correlates with effector function and is relevant in viral infections (HIV, EBV, CMV).
• Ligands: MHC Class I molecules (HLA-G, A, F, B27, E, F).
• Functional observation: HP-F1 reduces CD16-dependent cytolytic activity of NK cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated for flow cytometry of normal human peripheral blood cells.
Use at 5 µL (0.25 µg) per test in a final volume of 100 µL.
Cell number: 10⁵–10⁸ cells/test (empirically determined).
Excitation: 633–647 nm; Emission: 660 nm; Laser: Red Laser.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

This gene belongs to the leukocyte immunoglobulin-like receptor (LIR) family, located at chromosome 19q13.4.
The encoded protein is a subfamily B class receptor containing 2–4 extracellular immunoglobulin domains, a transmembrane domain, and 2–4 ITIM motifs.

• Expression site: Immune cells binding to MHC class I molecules on antigen-presenting cells.
• Function: Transduces inhibitory signals to regulate immune activation and prevent autoreactivity.
• Biological role: Controls inflammatory responses and cytotoxicity.
• Variants: Multiple transcript variants encoding different isoforms are known.
