Target Information
Mannose (or mannan)-binding lectin (MBL), also known as serum mannose-binding protein (MBP), initiates the lectin branch of the innate immune response by binding to the surface of potentially pathogenic microorganisms and initiating complement fixation through an N-terminal collagen-like domain.
MBL은 항체 비의존적 기전으로 병원체를 직접 중화할 수 있으며, MBL 유전자의 돌연변이는 보체 활성 저하로 인한 면역결핍과 관련이 있습니다.
인간에서 MBL과 결합하여 렉틴 경로를 활성화하는 세 종류의 MBL-관련 세린 프로테아제(MASP-1, MASP-2, MASP-3)와 MASP-2의 절단형(sMAP 또는 MAp19)이 존재합니다. 활성화된 MASP는 보체 경로의 하위 구성 요소를 절단 및 활성화합니다.
MASP-3는 MASP-1 유전자의 대체 스플라이싱 산물로, MBL 결합 경쟁을 통해 MASP-2의 활성화를 억제할 수 있습니다.
