Target Information

Glutamate is the main excitatory neurotransmitter in the brain. Traditionally, it was known to act on ligand-gated receptor channels such as NMDA, AMPA, and kainate receptors, which mediate fast excitatory synaptic transmission. More recently, glutamate has been shown to regulate enzymes producing second messengers via specific G-protein-coupled receptors known as metabotropic glutamate receptors (mGluRs).
Group-I receptors (mGluR1a and mGluR5) stimulate phospholipase C, increasing phosphoinositide turnover and calcium release from internal stores. Group-II (mGluR2, mGluR3) and Group-III receptors inhibit adenylyl cyclase activity.
