Product Specific Information

The eBioCB-CALLA monoclonal antibody recognizes human CD10 (CALLA, NEP, enkephalinase, Neprilysin), a 100 kDa type II cell surface glycoprotein originally identified on most acute lymphoblastic leukemias (ALL). CD10 is a Zn²⁺-dependent metallo-peptidase involved in regulation of chemotactic and inflammatory processes involving neutrophils. It also regulates stromal cell-dependent B lymphopoiesis and is expressed in mature B cells, enterocytes, renal tubules, and glomeruli.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on human peripheral blood cells. Recommended use: 5 µL (0.2 µg) per test in 100 µL final volume (10⁵–10⁸ cells/test).

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.

• Add 5 µL per sample (regardless of the number of NovaFluors).
• For single-color controls: add 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescence with minimal spillover and spread.

Target Information

CD10 (Common Acute Lymphocytic Leukemia Antigen, CALLA) is a cell surface enzyme with neutral metalloendopeptidase activity that inactivates various biologically active peptides. It is expressed on lymphoblastic, Burkitt’s, and follicular germinal center lymphomas, immature B cells, and cells from chronic myelocytic leukemia (CML) patients. CD10 is also found on breast myoepithelial cells, bile canaliculi, fibroblasts, and highly expressed in kidney and gut epithelial brush borders.

CD10 is a 100 kDa type II transmembrane glycoprotein encoded by a >45 kb gene with four alternatively spliced mRNA transcripts. Associated diseases include spinocerebellar ataxia 43 and Charcot-Marie-Tooth disease.
