Product Specific Information

The eBioCB-CALLA monoclonal antibody recognizes human CD10 (CALLA, NEP, enkephalinase, Neprilysin), a 100 kDa type II cell surface glycoprotein expressed in most acute lymphoblastic leukemias (ALL). CD10 is a Zn²⁺-dependent metallo-peptidase involved in regulating chemotactic and inflammatory processes, and stromal cell-dependent B lymphopoiesis. Expression is also found on enterocytes and renal tubules.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.2 µg)/test.
Cell number range: 10⁵–10⁸ cells/test.

Compatibility Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

CellBlox Plus Blocking Buffer:

• Required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample.

Target Information

CD10 (Common Acute Lymphocytic Leukemia Antigen, CALLA) is a cell surface enzyme with neutral metalloendopeptidase activity that inactivates various biologically active peptides. It is expressed on lymphoblastic, Burkitt’s, and follicular germinal center lymphomas, immature B cells, and chronic myelocytic leukemia (CML) cells. Also found on breast myoepithelial cells, bile canaliculi, fibroblasts, and kidney/gut epithelial brush borders.

CD10 is a 100 kDa type II transmembrane glycoprotein encoded by a >45 kb gene with four alternatively spliced mRNA transcripts. Associated diseases include spinocerebellar ataxia 43 and Charcot-Marie-Tooth disease.
