Product Specific Information

The eBioCB-CALLA monoclonal antibody recognizes human CD10 (CALLA, NEP, enkephalinase, Neprilysin), a 100 kDa type II cell surface glycoprotein originally identified in acute lymphoblastic leukemias (ALL). CD10 is a Zn²⁺-dependent metallo-peptidase involved in the regulation of chemotactic and inflammatory processes and stromal cell-dependent B lymphopoiesis. Expression is also found in enterocytes, renal tubules, and glomeruli.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells. Use 5 µL (0.2 µg) per test in a 100 µL final volume. Cell number range: 10⁵–10⁸ cells/test.

Note:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD10 (CALLA) is a cell surface enzyme with neutral metalloendopeptidase activity that inactivates various biologically active peptides. It is expressed on lymphoblastic, Burkitt’s, and follicular germinal center lymphomas, immature B cells, and chronic myelocytic leukemia (CML) cells. CD10 is also present on breast myoepithelial cells, bile canaliculi, fibroblasts, and highly expressed on kidney and gut epithelial brush borders.

CD10 cleaves peptides at the amino side of hydrophobic residues, inactivating peptide hormones such as glucagon, enkephalins, substance P, neurotensin, oxytocin, and bradykinin. It is a 100 kDa type II transmembrane glycoprotein encoded by a gene with multiple alternatively spliced 5′ untranslated regions.
