Target Information
The munc-18 interacting protein (Mint) family comprises evolutionarily conserved adaptor proteins involved in membrane transport and organization. Mammals express three isoforms: Mint1, Mint2, and Mint3. While the N-terminal region shows little sequence conservation, the C-terminal region is highly conserved and contains phosphotyrosine-binding (PTB) and PSD-95/DLG-A/ZO-1 (PDZ) domains, which serve as protein interaction modules.
Mint1 and Mint2 are expressed mainly in the brain and interact with Munc18, a key component of synaptic vesicle fusion. Mint3, by contrast, is ubiquitously expressed, with lowest levels in brain and testis, and does not interact with Munc18. Mint3 interacts with the Alzheimer’s disease-related amyloid precursor protein (APP) through its PTB and PDZ domains, potentially linking APP to transport machinery components and regulating its trafficking, endocytosis, and metabolism. Abnormal APP metabolism is associated with early-onset Alzheimer’s disease.
