The LG.3A10 monoclonal antibody reacts with mouse CD27, a lymphocyte-specific member of the TNFR superfamily. CD27 is expressed by virtually all mature T cells and by a subpopulation of B cells, mainly memory B cells. In mouse, CD27 is found on nearly all thymocytes except a population of CD46-CD8⁻ precursors. CD27 binds to CD70 and plays an important role in T cell–B cell interaction and effector cell maturation.

When crosslinked on purified T cells, LG.3A10 increases proliferation. Without crosslinking, it inhibits CD27–CD70 interaction.

Applications Reported: Flow cytometric analysis and immunohistology staining of frozen tissue sections.
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes at ≤0.25 µg/test (100 µL final volume). Cell number should be empirically determined (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.
Storage and Handling: Use in a sterile environment.

Target Information

CD27 is a 50 kDa member of the tumor necrosis factor (TNF) receptor superfamily, including CD40 and CD30. It regulates cell proliferation and death and is mainly expressed on lymphocytes. CD27 forms a disulfide-linked homodimer on mature thymocytes, peripheral blood T cells, and a subset of B cells. Activation of T cells via TCR-CD3 complex increases CD27 expression and releases soluble CD27 (sCD27), a prognostic marker for B cell malignancies.

CD27-binding protein (SIVA) is a proapoptotic protein involved in CD27-mediated apoptosis. Dysfunctions in CD27 are associated with Lymphoproliferative Syndrome 2 and Autosomal Recessive Lymphoproliferative Syndrome.
