Product Specific Information

Description:
The eBio3C7 antibody reacts with mouse CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Ra). CD25 is expressed by early progenitors of the T and B lineage as well as by activated mature T and B lymphocytes. CD25 binds IL-2 with low affinity alone but forms a high-affinity receptor when associated with CD122 (IL-2Rβ) and CD132 (IL-2Rγ). Binding of IL-2 to both receptor classes is inhibited by the 3C7 antibody. CD25 plays a role in lymphocyte differentiation, activation, and proliferation. The epitopes recognized by eBio3C7, PC61.5, and eBio7D4 are distinct, allowing depletion with one antibody and detection with another.

Applications Tested:
Validated by flow cytometric analysis of stimulated mouse splenocytes. Recommended ≤ 0.5 µg/test (100 µL final volume). Optimal titration advised for best assay performance. Fluorochrome-conjugated eBio3C7 (3C7) is recommended for flow cytometry.

Target Information

CD25 (IL2 receptor alpha chain, IL2RA) is a cytokine receptor involved in T and B lymphocyte proliferation. The high-affinity IL-2 receptor complex comprises IL2RA, IL2RB, and IL2RG. Homodimeric IL2RA forms a low-affinity receptor, while IL2RB homodimers yield medium affinity. IL2 expression in mature thymocytes is monoallelic, serving as a regulatory mechanism for gene expression. IL2, produced mainly by mature T cells, functions as a growth factor, differentiation factor, and apoptosis regulator. It enhances B cell proliferation, NK cell activity, and inhibits granulocyte macrophage colony formation. Disruption of IL2RA in mice results in ulcerative colitis-like disease, implicating its role in immune response regulation. Mutations are linked to interleukin-2 receptor alpha deficiency.
