Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human IL-10. Anti-Human IL-10-specific antibody was purified by affinity chromatography and then biotinylated.

Sandwich ELISA:
To detect hIL-10 by sandwich ELISA (100 µL/well antibody solution), use 0.25–1.0 µg/mL. Works with PeproTech Polyclonal Anti-Human IL-10 (500-P20) as capture antibody, detecting at least 0.2–0.4 ng/well of Recombinant hIL-10.

Western Blot:
For detecting hIL-10 by Western blot, use 0.1–0.2 µg/mL. Detection limit: 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information

IL-10 encodes a cytokine primarily produced by monocytes, with additional production by lymphocytes. It plays a key role in immunoregulation and inflammation by:

• Down-regulating Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages
• Enhancing B cell survival, proliferation, and antibody production
• Blocking NF-kappa B activity and regulating the JAK-STAT signaling pathway

Knockout studies in mice indicate IL-10 is essential for intestinal immune regulation. Mutations are linked to higher susceptibility to HIV-1 infection and rheumatoid arthritis.
Associated diseases include Graft-Versus-Host Disease and Human Immunodeficiency Virus Type 1.
