- Paraffin section staining requires pre-treatment.
- For tissues fixed <24 hrs: use Proteinase K.
- For tissues fixed >24 hrs: use Citrate buffer (pH 6.0).
- For FACS analysis: use 10 µL of working dilution to label 1×10⁶ cells in 100 µL.
- Rat anti-mouse F4/80 antibody (Clone Cl:A3-1) recognizes murine F4/80 antigen (~160 kDa), a cell surface glycoprotein of the EGF-TM7 family sharing 68% amino acid identity with human EMR1.
Target Information
F4/80 antigen is a 160 kDa cell surface glycoprotein, member of the EGF TM7 family. It is expressed on a wide range of mature tissue macrophages (Kupffer cells, Langerhans, microglia, gut lamina propria, peritoneal cavity, lung, thymus, bone marrow stroma, spleen red pulp). Expression varies during macrophage maturation and activation. Reported on a subpopulation of dendritic cells, absent in macrophages of T cell areas in spleen and lymph node. Biological functions are not fully defined, but may play a role in generation of efferent CD8⁺ regulatory T cells.
