This antibody detects endogenous levels of FOXO1A only when phosphorylated at Serine 319.
Target Information
FOXO1 (FKHR, ForKHead Box O1) is a member of the forkhead homeotic gene family of transcription factors. It can act as a coactivator or corepressor of nuclear receptor activity mediated through the LXXLL motif in the carboxyl terminal region. Association with PAX3 has been implicated in alveolar rhabdomyosarcoma. FOXO1 is linked to anti-tumor activity, autophagy, and myogenic differentiation. It is a key target of insulin signaling regulating metabolic homeostasis under oxidative stress. FOXO1 binds to the insulin response element (IRE) and Daf-16 family binding element (DBE) to regulate gene expression involved in redox balance, osteoblast regulation, and glucose metabolism. In hepatocytes, FOXO1 promotes gluconeogenesis in cooperation with PPARGC1A by activating genes such as IGFBP1, G6PC, and PPCK1. It also acts downstream of CDK1, PKB/AKT1, and SKT4/MST1 to regulate cell death.
