This antibody detects endogenous levels of FOXO1A only when phosphorylated at Serine 256.
Target Information
FOXO1 (FKHR, Forkhead Box O1) is a transcription factor belonging to the forkhead homeotic gene family. It can act as a coactivator or corepressor of nuclear receptor activity through the LXXLL motif in its C-terminal region. FOXO1 is associated with PAX3 in alveolar rhabdomyosarcoma and is linked to autophagy, myogenic growth, and differentiation. It is a key target of insulin signaling that regulates metabolic homeostasis under oxidative stress. FOXO1 binds to the insulin response element (IRE) and the Daf-16 family binding element (DBE) to control redox balance, osteoblast numbers, and bone mass. It also regulates glucose metabolism and suppresses RUNX2 activity. In hepatocytes, FOXO1 promotes gluconeogenesis by activating genes such as IGFBP1, G6PC, and PCK1. It is an important regulator of cell death downstream of CDK1, PKB/AKT1, and MST1.
