Product Specific Information

eBioTY/11.8 recognizes CD73, a 69-kDa GPI-anchored cell-surface protein with ecto-5'-nucleotidase activity.
Expression on myeloid cells (CD11b) is restricted to the bone marrow. In human cells, CD73 can be induced to secrete a soluble form with IL-2, suggesting a role in mediating activation signals.

CD73 is expressed on a subset of lymphocytes and increases during lymphocyte maturation. Memory CD4 T cells expressing CD73 are similar to uncommitted primed precursor helper cells (Thpp) that can differentiate into Th1 or Th2 cells. CD73 is also found on regulatory T cells.

Each product contains:

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes, ≤0.125 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
Careful titration is recommended for optimal performance.

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor dyes.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which enhances staining performance.
Add 5 µL per sample to antibody cocktails or single-color controls (100 µL cell sample, 10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, providing engineered fluorescent signatures optimized for spillover and spread.

Internal testing indicates NovaFluor Yellow 730 may non-specifically stain B cells in SJL mice, but not in BALB/c or C57BL/6 strains.

Target Information

CD73 (Ecto-5'-nucleotidase) catalyzes the conversion of purine 5'-mononucleotides to nucleosides, primarily AMP to adenosine, at neutral pH.
It forms a dimer of two identical 70 kDa subunits linked to the plasma membrane via GPI.
CD73 serves as a marker of lymphocyte differentiation; deficiencies are linked to immunodeficiency diseases.
The CD73 gene is located at 6q14–q21.
Defects can lead to joint and arterial calcification or intestinal tuberculosis.
Two transcript variants encoding different isoforms are known.
