Product Specific Information

The B20.1 monoclonal antibody reacts with mouse T-cell receptor (TCR) V alpha 2 chains, recognizing the majority of the TCR V alpha 2 subfamily in mice carrying the a, b, and c haplotypes. It also reacts with V delta 8 due to high homology.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse lymph node cells (≤0.125 µg/test)

A test is defined as staining a cell sample in 100 µL final volume. Cell number may range from 10⁵ to 10⁸ cells/test.
Preblock Fc receptors for 10 minutes using 0.5 µg of purified anti-mouse CD16/32 (Product #14-0161-82).
Carefully titrate antibody for optimal performance.

Compatibility Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

Blocking Buffer Usage:

• CellBlox Plus Blocking Buffer (Product #C001T06F01) is required for optimal staining.
• Add 5 µL per sample (regardless of number of NovaFluors).
• For single-color controls: use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology using novel nucleic acid dye structures for engineered fluorescent signatures, minimizing spillover and spread.

Target Information

T-cell receptors (TCR) recognize peptides presented by MHC class II molecules, essential for adaptive immune response. TCR recognition of self-peptides is linked to autoimmune disease and tumor antigens. Superantigen engagement can cause toxic shock syndrome.

Autoantibodies to TCR V beta segments are found in RA and SLE, modulating inflammatory responses. Most human T cells express TCR alpha-beta with CD4 or CD8; a small subset (DN T cells) lacks both markers and may increase in autoimmune or immunodeficiency disorders. Gamma-delta T cells, found mainly in epithelium, exhibit unique antigen recognition and immunoregulatory activity.
