Product Specific Information

Description:
eBioTY/11.8 recognizes CD73, a 69-kDa GPI-anchored cell-surface protein with ecto-5’-nucleotidase activity. Expression on myeloid cells (CD11b) is restricted to the bone marrow. In humans, CD73 can be induced to secrete a soluble form with IL-2, suggesting a role in mediating activation signals. Differences between human and mouse CD73 have been reported.

CD73 is expressed on a subset of lymphocytes and increases during lymphocyte maturation. It is found on memory CD4 T cells and regulatory T cells.

Each product contains 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported:
This eBioTY/11.8 (TY/11.8) antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended use ≤0.125 µg per test. A test is defined as the amount of antibody that stains a cell sample in a final volume of 100 µL. Cell number should be determined empirically (10⁵–10⁸ cells/test).

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
    • Add 5 µL per sample (regardless of number of NovaFluors).
    • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Strain-specific Note:
NovaFluor Yellow 690 non-specifically stains B cells in SJL mice. Non-specific staining not observed in BALB/c or C57BL/6 mice.

NovaFluor conjugates are based on Phiton™ technology utilizing nucleic acid dye structures for engineered fluorescent signatures with minimal spillover and spread.

Target Information

CD73 (Ecto-5'-nucleotidase) catalyzes the conversion of purine 5'-mononucleotides to nucleosides, preferring AMP as substrate. It is a dimer (70 kDa subunits) anchored to the plasma membrane via GPI linkage. CD73 serves as a marker of lymphocyte differentiation, and deficiency is associated with immunodeficiency diseases. The CD73 gene is localized to 6q14–q21. Defects can lead to calcification of joints and arteries, and intestinal tuberculosis. Two transcript variants encoding different isoforms have been identified.
