Description:
The N2B2 monoclonal antibody reacts with mouse TNF-related apoptosis-inducing ligand (TRAIL), a member of the TNF superfamily. TRAIL is not detected on freshly isolated T, B, or NK cells but can be induced on CD3^- NK1.1^+ NK cells after IL-2 or IL-15 stimulation. N2B2 inhibits IL-2- or IL-15-activated NK cell cytotoxicity against mouse fibrosarcoma L929 target cells.

Applications Reported:
N2B2 has been reported for use in flow cytometric analysis and blocking of TRAIL in functional assays.

Applications Tested:
The N2B2 antibody has been tested by flow cytometric analysis of mouse TRAIL transfected cells and can be used at ≤1 µg per test (100 µL final volume). Cell number should be determined empirically (10⁵–10⁸ cells/test). Titrate for optimal performance.

Target Information

TRAIL is a cytokine belonging to the TNF ligand family. It preferentially induces apoptosis in transformed and tumor cells while sparing normal cells. TRAIL binds to multiple TNF receptor superfamily members including TRAILR1 (DR4), TRAILR2 (DR5), TRAILR3 (DcR1), TRAILR4 (DcR2), and OPG. Binding to decoy receptors (TRAILR3, TRAILR4, OPG) modulates activity without triggering apoptosis. TRAIL signaling activates MAPK8/JNK, caspase 8, and caspase 3 pathways. It is a type II membrane protein expressed in various human tissues and induces apoptosis and NF-κB activation similar to TNF and Fas ligand.
