Product Specific Information

Description:
The 188 monoclonal antibody was generated against and reacts with mouse B7-H4 (also known as B7S1, B7X). Cross-reactivity with other proteins has not been determined. B7-H4 is a member of the B7 family that inhibits T-cell activation, cell cycle progression, and IL-2 production. The ligand for B7-H4 has not been identified. Double staining with antibodies Clone 9 and 188 indicates distinct epitopes or no steric hindrance. Clone 188 stains mouse B7-H4 transfected cells but not spleen cells. The expression pattern of B7-H4 remains incompletely characterized.

Applications Reported:
This antibody is reported for use in flow cytometric analysis and for blocking B7-H4 ligand binding in vitro.

Applications Tested:
Tested by flow cytometric analysis of transfected cells. Recommended ≤0.5 µg per test (100 µL final volume). Cell number may vary (10⁵–10⁸ cells/test). Titrate for optimal performance.

Storage and Handling:
Use in a sterile environment.

Filtration:
0.2 µm post-manufacturing filtered.

Purity:

90%, determined by SDS-PAGE.

Target Information

B7-H4 (B7x, VTCN1):
A costimulatory protein functioning as a negative regulator of T-cell mediated immunity. Although its receptor is unknown, B7-H4 delivers inhibitory signals preventing T-cell proliferation, cell cycle progression, and IL-2 production. It is expressed on activated T-cells, B-cells, monocytes, and dendritic cells. Aberrant expression is linked to lung, breast, and ovarian cancers. Overexpression may suppress T-cell immunity and promote tumor progression. VTCN1 may serve as a biomarker for early detection of ovarian cancer and renal cell carcinoma (RCC).
