Product Specific Information

The LH1 monoclonal antibody reacts with mouse Herpes Virus Entry Mediator (HVEM, TR2), a member of the TNF-receptor superfamily. HVEM is expressed on various cell types, including T cells, B cells, monocytes, neutrophils, and dendritic cells. It functions as a cellular mediator for herpes simplex virus (HSV) entry via binding with viral envelope glycoprotein D (gD). The cytoplasmic region of HVEM interacts with TRAF family members, mediating immune response signaling pathways.

HVEM also serves as a ligand for BTLA (B and T lymphocyte attenuator), indicating its role in lymphocyte activation and immune homeostasis. The LH1 antibody has been reported as a blocking antibody that interferes with the HVEM-LIGHT interaction but not with HVEM-BTLA binding.

Applications Reported: Functional assays and flow cytometric analysis (fluorochrome-conjugated LH1 recommended for flow cytometry).
Applications Tested: Validated for flow cytometric analysis of mouse splenocytes. Recommended usage ≤0.25 µg per test (100 µL final volume, 10⁵–10⁸ cells/test). Optimal titration is advised for specific assays.

Storage and Handling: Use in a sterile environment.
Filtration: 0.2 µm post-manufacturing filtered.
Purity: >90% (SDS-PAGE).
Endotoxin Level: <0.001 ng/µg antibody (LAL assay).

Target Information

Gene: TNFRSF14 (HVEM)
Function: Member of the TNF-receptor superfamily; mediates HSV entry via glycoprotein D binding. The cytoplasmic region binds TRAF family members, activating immune signaling pathways.
Biological Role: Activation of TNFRSF14 in T cells stimulates proliferation and cytokine production, enhancing CTL-mediated tumor immunity. Multiple alternatively spliced transcript variants have been reported.
