Product Specific Information
The SHM33 monoclonal antibody reacts with human CD323, also known as Junctional Adhesion Molecule (JAM) 3. CD323 is a member of the Ig superfamily of transmembrane proteins and is expressed on endothelial and epithelial cells, smooth muscle cells, peripheral nerves, platelets, and some B cells. It is also expressed on VEGF and histamine-stimulated endothelial cells and activated T cells.
CD323 plays roles in neutrophil transmigration during inflammation, angiogenesis, cell polarity, and nerve conduction. It interacts homo- and heterotypically with beta2 and beta3 integrins, JAM2, and the viral receptor CAR.
Applications Reported: Suitable for microscopy, immunohistochemical staining, and immunocytochemistry.
Applications Tested: Tested by immunocytochemistry of methanol-fixed human cells; usable at ≤1 µg/mL. Optimize titration for best results.
Filter Recommendation: Use filters capturing 570 nm emission (e.g., Excitation 545/25, 565LP, Emission 605/70). Standard Alexa Fluor® 555 or TRITC filters are acceptable.

Target Information
JAM3 is involved in tight junction formation and cell-to-cell adhesion in epithelial or endothelial sheets, forming continuous seals that serve as physical barriers to solute and water passage. The encoded protein localizes in tight junctions between high endothelial cells. Unlike other members of this family, JAM3 does not adhere to leukocyte cell lines and forms weak homotypic interactions. It acts as a receptor for another junctional adhesion molecule family member.
