Description:
The RA3-6B2 monoclonal antibody reacts with the exon A-restricted isoform of mouse CD45, a 220 kDa surface molecule. CD45R/B220 epitope is primarily expressed by B cell lineage from early Pro-B to mature B cells. Some activated T cells, LAK cells, NK progenitors, and T cells of lpr/lpr mutant mice also express this antigen.

Applications Reported:
Reported for use in microscopy, immunocytochemistry, and immunohistochemistry (formalin-fixed paraffin-embedded or frozen tissue sections).

Applications Tested:
Tested by IHC of frozen mouse spleen; effective at ≤1 µg/mL. Optimal dilution should be determined experimentally.

Filter Recommendation:
For eFluor™ 570 conjugate, use filters capturing 570 nm emission (e.g., Excitation 545/25, 565LP, Emission 605/70). Standard Alexa Fluor™ 555 or TRITC filters are acceptable.

Target Information

CD45R (PTPRC) is a member of the protein tyrosine phosphatase (PTP) family, regulating various cellular processes such as cell growth, differentiation, and signal transduction. It is expressed in hematopoietic cells and is essential for T- and B-cell receptor signaling. CD45 glycoprotein plays a key role in lymphocyte development and antigen signaling, acting as a regulator of Src-family kinases. Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions. CD45 also modulates integrin-mediated signaling and apoptosis-related DNA fragmentation. CD45RA is a restricted isoform expressed in specific lymphoid subtypes. Dysfunction of CD45 is associated with Severe Combined Immunodeficiency and Hepatitis C.
