Product Specific Information

Description:
The Michel-17 monoclonal antibody reacts with mouse CD83, a 45 kDa cell surface glycoprotein and a member of the Ig superfamily. CD83 is predominantly expressed on mature dendritic cells (DCs) and activated lymphocytes. Cross-linking of CD83 with Michel-17 on DCs or activated T cells does not induce activation signals. CD83 plays an important role in T cell development through ligand interaction. CD83-Ig protein studies have shown a CD83 ligand mainly expressed by B220⁺ cells in mouse spleen.

  • Recommended ≤0.5 µg per test (100 µL final volume).
  • Optimal cell number: 10⁵–10⁸ cells/test (empirically determined).
  • Antibody should be titrated for optimal assay performance.

Target Information

CD83 is a 40–45 kDa cell surface glycoprotein expressed by peripheral blood dendritic cells. Peripheral lymphocytes can express low levels of CD83 when cultured with agents such as Con A or PHA. In immunohistology, CD83 is strongly expressed by interfollicular interdigitating reticulum cells and weakly by cells within germinal centers. It is also expressed by Langerhans cells in the skin.

The CD83 antigen is a 186-amino-acid single-chain glycoprotein, part of the immunoglobulin superfamily, consisting of an extracellular V-type Ig-like domain, a transmembrane region, and a short cytoplasmic tail (~40 amino acids). CD83 undergoes extensive post-translational glycosylation, resulting in a molecular weight approximately twice that of its core protein. CD83⁺ cells exhibit a unique surface phenotype distinct from T, B, NK, or myelomonocytic lineages, co-expressing high levels of MHC class II molecules and various lineage markers (CD2, CD5, CD40, CD78, CD13, CD33, CD36).

Diseases associated with CD83 dysfunction include plague and Rift Valley Fever.
