Antibody detects endogenous levels of MST1 when phosphorylated at Thr183 or MST2 when phosphorylated at Thr180.


Target Information

STK3 and STK4 are stress-activated, pro-apoptotic kinases that, following caspase cleavage, enter the nucleus and induce chromatin condensation and DNA fragmentation. They are key components of the Hippo signaling pathway, which regulates organ size and tumor suppression by limiting proliferation and promoting apoptosis.

The pathway involves a kinase cascade where STK3/MST2 and STK4/MST1, in complex with SAV1, phosphorylate and activate LATS1/2 with MOB1, which then phosphorylates and inactivates YAP1 and WWTR1/TAZ. Phosphorylation of YAP1 prevents its nuclear translocation, regulating genes involved in proliferation, cell death, and migration.

STK3/MST2 and STK4/MST1 repress hepatocyte proliferation, prevent activation of adult liver stem cells, and inhibit tumor formation. They phosphorylate NKX2-1, NEK2, MOBKL1A, MOBKL1B, and RASSF2, and regulate centrosome disjunction and ESR1 transcriptional activity. They also positively regulate RAF1 activation by suppressing inhibitory phosphorylation on Ser-259.
