Product Specific Information
The Ber-H2 monoclonal antibody reacts with human CD30, a 120 kDa transmembrane glycoprotein of the TNFR superfamily. CD30 is expressed on activated T and B lymphocytes, but not on resting lymphocytes. It is also found on certain lymphomas, embryonal carcinomas, and virus-infected lymphocytes (HIV, HTLV, EBV). Interaction with its ligand CD30L leads to activation of NF-kappaB and regulation of apoptosis.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometry on stimulated human peripheral blood cells at 5 µL (0.06 µg)/test.
A test corresponds to staining a 100 µL cell sample (10^5–10^8 cells/test).
Excitation: 633–647 nm
Emission: 660 nm
Laser: Red Laser
Target Information
CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein originally identified on Hodgkin’s and Reed-Sternberg cells. CD30L (CD153) is its ligand, mediating cell proliferation, activation, differentiation, and apoptosis. It plays a key role in Hodgkin’s disease and other CD30+ lymphomas, acting as a costimulatory molecule in thymic negative selection. CD30 expression occurs on activated T and B cells, virus-infected lymphocytes, and some non-Hodgkin’s lymphomas. The soluble form (sCD30) reflects Th2 immune response. TRAF2 and TRAF5 interact with CD30 to activate NF-kappaB signaling. CD30 is a positive regulator of apoptosis and limits autoreactive CD8 effector T cell proliferation. Two alternatively spliced transcript variants exist. CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, and most Anaplastic Large Cell Lymphomas (ALCL), as well as embryonal carcinomas.
