This antibody binds to carboxy-terminal oligo-histidine tags (6x His tags) of fusion proteins. It also has affinity for internal His-tags but no affinity for N-terminal His-tags.

Target Information

The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH). The His tag is commonly expressed at either the N- or C-terminal regions of recombinant proteins to allow isolation or purification by immobilized metal affinity chromatography (IMAC).

Epitope tagging enables detection of recombinant proteins for which no specific antibody exists. His-tagged proteins, containing histidine clusters at the carboxyl or amino terminus, can be purified by metal chelate chromatography due to the strong, selective chelation between histidine residues and Ni²⁺ ions (e.g., Ni²⁺-NTA adsorbent). His tag-specific antibodies facilitate detection or co-immunoprecipitation of tagged proteins.
