이 항체는 융합 단백질의 카복시 말단 6x-His 태그(oligo-histidine tags)에 결합합니다. 내부 His-tag에도 결합하지만 N-말단 His-tag에는 결합하지 않습니다.
Target Information
6x His 태그는 6개의 연속된 히스티딘 잔기(HHHHHH)로 구성된 합성 올리고 펩타이드입니다. 일반적으로 재조합 단백질의 N-말단 또는 C-말단에 부착되어 금속 친화성 크로마토그래피(IMAC)를 이용한 단백질 정제에 사용됩니다.
Epitope tagging은 알려진 에피토프를 재조합 단백질에 융합하여 항체가 없는 단백질도 검출할 수 있게 하는 기술입니다.
히스티딘 태그 단백질은 금속 킬레이트 크로마토그래피를 통해 정제되며, 특히 Ni²⁺-NTA 흡착체를 이용한 친화 크로마토그래피에서 높은 선택성과 친화력을 보입니다.
His tag 특이적 항체는 태그 단백질의 검출 및 공동면역침전(Co-IP)에 활용됩니다.
