Target Information
CHEK1 (CHK1) is a serine/threonine protein kinase that prevents replication when DNA integrity is compromised. It acts as a key mediator in the DNA damage-induced checkpoint network and is evolutionarily conserved.
Inhibition of CHK1 can abrogate the G2 or S checkpoint, leading to mitotic catastrophe and apoptosis in p53-deficient cancer cells, while normal cells remain arrested in G1.
Therefore, CHK1 inhibitors can enhance the efficacy of DNA-damaging agents and are promising targets for cancer therapy, especially for p53-deficient tumors.
