Product Specific Information

The J105 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the CD28 immunoglobulin superfamily. PD-1 contains an immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a critical role in peripheral tolerance and autoimmune disease. PD-1 is expressed predominantly on activated T and B lymphocytes.

PD-1 ligands include PD-L1 (B7-H1) and PD-L2 (B7-DC), which exhibit overlapping functions. These ligands are constitutively expressed on some normal tissues and upregulated on activated antigen-presenting cells.

Costaining experiments suggest that eBioJ105 recognizes a different epitope than MIH4 (Product #11-9969).

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on stimulated normal human peripheral blood cells. Recommended usage is 5 µL (0.5 µg) per test (100 µL final volume). Cell count may range from 10⁵ to 10⁸ cells/test.

Light Sensitivity: Protect from light due to photo-induced oxidation sensitivity.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark.

Target Information

T-cell activation is self-limited by inhibitory receptors such as PD-1 (PDCD-1), which mediate negative signals upon binding to PD-L1 or PD-L2 on antigen-presenting cells. These interactions inhibit immune activation in the absence of danger signals such as LPS. PD-1 deficiency leads to hyperactivated immune responses and autoimmune phenotypes. Despite its predicted molecular weight, PD-1 often migrates at higher molecular weight in SDS-PAGE.
