Target Information
CD20 is a non-glycosylated surface phosphoprotein with a molecular weight of 33–37 kDa depending on phosphorylation. It is expressed on mature and most malignant B cells, as well as a subpopulation of T lymphocytes and follicular dendritic cells. CD20 expression correlates with surface IgM expression and plays roles in calcium conductance, cell cycle progression, and B-cell proliferation.
CD20 is associated with lipid rafts, and its interaction depends on extracellular triggers and BCR aggregation. Upon receptor ligation, CD20 and BCR colocalize transiently before BCR endocytosis, while CD20 remains on the cell surface. Because of its stable surface expression, CD20 is a valuable target for antibody-mediated B-cell depletion therapies. CD20 dysfunction is linked to Ms4a1-related common variable immune deficiency.
