Target Information
CD20 is a non-glycosylated surface phosphoprotein with a molecular weight of 33–37 kDa depending on phosphorylation. It is expressed on mature and most malignant B cells, as well as on a subset of T lymphocytes and follicular dendritic cells. CD20 expression correlates with surface IgM expression and plays a role in calcium conductance, cell cycle progression, and B-cell proliferation.
CD20 is associated with lipid rafts, and its distribution is influenced by extracellular signals and B-cell receptor (BCR) aggregation. Following receptor ligation, CD20 and BCR colocalize and then dissociate before BCR endocytosis, with CD20 remaining on the cell surface. Because of its stable surface expression, CD20 serves as an effective target for antibody-mediated B-cell depletion in research on B-cell malignancies.
Diseases associated with CD20 dysfunction include MS4A1-related common variable immune deficiency.
