Target Information
CD20 is a non-glycosylated surface phosphoprotein with a molecular weight of 33–37 kDa depending on phosphorylation. It is expressed on mature and most malignant B cells, as well as on a subset of T lymphocytes and follicular dendritic cells. CD20 expression coincides with surface IgM expression and plays a role in calcium conductance, cell cycle progression, and B-cell proliferation.
CD20 associates with lipid rafts, and its conformation changes upon extracellular triggering or BCR aggregation. After receptor ligation, CD20 and BCR colocalize but rapidly dissociate before BCR endocytosis, while CD20 remains on the cell surface. Due to its stable surface expression, CD20 is a valuable target for antibody-mediated therapeutic depletion of B cells, particularly in B-cell malignancies. Dysfunctions of CD20 are linked to Ms4a1-related common variable immune deficiency.
