Target Information
CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa) expressed on mature and most malignant B cells, as well as subsets of T lymphocytes and follicular dendritic cells.
It regulates transmembrane calcium conductance, cell cycle progression, and B-cell proliferation.
CD20 is associated with lipid rafts, with its interaction influenced by extracellular signaling and BCR aggregation.
Following receptor ligation, CD20 and BCR colocalize transiently before BCR endocytosis, while CD20 remains at the cell surface.
CD20 is an important target for antibody-mediated B-cell depletion therapies, as it is highly expressed on most B-cell malignancies and remains membrane-bound after antibody treatment.
Dysfunction of CD20 has been linked to Ms4a1-related common variable immune deficiency.
