Target Information

CD47 (integrin-associated protein, IAP) is a ubiquitously expressed transmembrane glycoprotein that interacts with integrins and regulates their functions. It modulates signaling pathways such as activation of heterotrimeric G proteins and increases intracellular calcium upon cell adhesion.
CD47 serves as a receptor for the C-terminal cell-binding domain of thrombospondin and plays roles in membrane transport and signal transduction. It is broadly distributed across tissues and reduced on Rh erythrocytes.
CD47 acts as a ligand for CD172a (SIRPα), an inhibitory receptor on macrophages, preventing phagocytosis of CD47-positive cells. The CD47-CD172a interaction influences cell migration, B cell adhesion, T cell activation, chondrocyte mechanotransduction, and neuronal development, particularly in synapse-rich regions of the brain and retina.
