Description:
eBioTY/11.8 recognizes CD73, a 69-kDa GPI-anchored cell-surface protein with ecto-5'-nucleotidase activity.
Expression on myeloid cells (CD11b⁺) is restricted to bone marrow. In humans, CD73 can be induced to secrete a soluble form with IL-2, suggesting a role in activation signaling. CD73 expression differs between species; for example, BALB/c mice have more CD4⁺CD73⁺ than CD8⁺CD73⁺ cells, while the reverse is observed in humans.

CD73 is expressed on subsets of lymphocytes and increases during lymphocyte maturation. It has been found on memory CD4 T cells and regulatory T cells, associated with Thpp precursor helper cells that can differentiate into Th1 or Th2 cells.

For flow cytometric analysis.

Applications Tested:
Validated in flow cytometric analysis of mouse splenocytes. Recommended usage ≤ 0.25 µg per test. Antibody should be titrated for optimal performance.

Fluorochrome Information:
Super Bright 436 is excited by the violet laser (405 nm) and emits at 436 nm. Use a 450/50 bandpass filter or equivalent. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

CD73 (Ecto-5'-nucleotidase, 5'-ribonucleotide phosphohydrolase):
Catalyzes conversion of purine 5'-mononucleotides to nucleosides at neutral pH, preferentially AMP.
Composed of two identical 70 kDa subunits bound to the plasma membrane via GPI linkage.
Acts as a marker of lymphocyte differentiation; deficiency linked to immunodeficiency diseases.
Other 5'-nucleotidase forms exist in cytoplasm and lysosomes, differing in substrate affinity and enzyme regulation.
CD73 gene localized to 6q14–q21; mutations may cause joint and arterial calcification or intestinal tuberculosis.
Two transcript variants encoding different isoforms have been identified.
