Target Information
FOXO1 (FKHR, Forkhead Box O1)는 transcription factor 계열의 forkhead homeotic gene family 하위군에 속합니다.
FOXO1은 LXXLL motif를 통해 핵 수용체 활성의 공동활성화(coactivator) 또는 공동억제(corepressor) 역할을 할 수 있습니다.
PAX3와의 결합은 alveolar rhabdomyosarcoma와 관련되어 있으며, FOXO1은 자가포식(autophagy), 근육 성장 및 분화, 항암 활성에 관여합니다.
또한 FOXO1은 인슐린 신호전달의 주요 표적이며, 산화 스트레스에 대한 대사 항상성을 조절합니다.
FOXO1은 인슐린 반응 요소(IRE) 및 Daf-16 결합 요소(DBE)에 결합하여 redox balance, 골형성, 골량 조절 및 골격의 내분비 기능을 조절합니다.
간세포에서 FOXO1은 PPARGC1A와 함께 IGFBP1, G6PC, PPCK1 등의 유전자 발현을 활성화하여 포도당 신생합성을 촉진합니다.
FOXO1은 CDK1, PKB/AKT1, SKT4/MST1 하류에서 세포사멸을 조절하는 중요한 인자입니다.
