Product Specific Information
The N2B2 monoclonal antibody reacts with mouse TNF-related apoptosis-inducing ligand (TRAIL), a member of the TNF superfamily. TRAIL is not detected on the surface of freshly isolated T, B, or NK cells, but can be induced preferentially on CD3⁻ NK1.1⁺ NK cells after stimulation with IL-2 or IL-15. N2B2 inhibits IL-2- or IL-15-activated NK cell cytotoxicity against mouse fibrosarcoma L929 target cells.
Applications Reported: N2B2 has been reported for use in flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse TRAIL transfected cells and can be used at ≤0.25 µg/test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.

Target Information
TRAIL is a cytokine belonging to the tumor necrosis factor (TNF) ligand family. It preferentially induces apoptosis in transformed and tumor cells but not in normal cells. TRAIL binds to multiple TNF receptor family members including TNFRSF10A (TRAILR1), TNFRSF10B (TRAILR2), TNFRSF10C (TRAILR3), TNFRSF10D (TRAILR4), and possibly TNFRSF11B (OPG).
Decoy receptors such as TRAILR3, TRAILR4, and OPG modulate its activity by preventing apoptosis induction. Binding of TRAIL to its receptors activates MAPK8/JNK, caspase 8, and caspase 3 pathways. Receptors include death domain-containing DR4 and DR5, and decoy receptors DcR1 and DcR2 lacking death domains. TRAIL is a type II membrane protein expressed in various human tissues and induces apoptosis and NF-κB activation.
