The BTLA antibody reacts with mouse BTLA (B and T lymphocyte attenuator) from both BALB/c and C57Bl/6 strains.
BTLA is expressed by peripheral lymphocytes, splenic macrophages, developing B cells in the bone marrow, and developing T cells in the thymus. It is also expressed in mature, but not immature, bone marrow-derived dendritic cells.
BTLA functions as a negative regulator of activation and/or function of various hematopoietic cell types. It has been reported to bind B7-H4, though further studies are needed to confirm this interaction.
Note: The anti-mouse BTLA monoclonal antibody 6F7 stains CD4+ and CD8+ single-positive thymocytes (Hurchla et al.), whereas clones 8F4, 3F9.D12, 6G3, and 6H6 do not. However, in splenocytes and bone marrow cells, these clones stain similar populations.
This 8F4 antibody has been tested by flow cytometric analysis of mouse splenocytes and can be used at ≤ 0.25 µg per test.
A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL.
Cell number should be determined empirically (typically 10⁵–10⁸ cells/test).
It is recommended to titrate the antibody for optimal assay performance.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
BTLA (B and T-lymphocyte attenuator) is a co-inhibitory receptor of the CD28 superfamily.
It acts as a lymphocyte inhibitory receptor that suppresses lymphocyte activation during immune responses.
BTLA is constitutively expressed on most CD4+ and CD8+ T cells, with expression decreasing upon activation.
It remains expressed on Th1 but not Th2 cells.
BTLA interacts with herpesvirus entry mediator (HVEM), a TNF receptor superfamily member, forming an important pathway regulating lymphocyte activation and immune homeostasis.
