Product Specific Information

The IK22/5 monoclonal antibody reacts with mouse CD254, also known as TRANCE (TNF-related activation-induced cytokine receptor), RANKL (receptor activator of NF-kappa B ligand), and OPGL (Osteoprotegerin Ligand).
TRANCE is a type II membrane protein (~35 kDa) closely related to TRAIL, FasL, and TNF. It is upregulated by TCR stimulation and expressed mainly by mouse T cells in thymus and lymph nodes.
TRANCE-deficient mice lack osteoclasts, indicating its critical role in osteoclast differentiation and T cell–dendritic cell interactions. The TRANCE–RANK interaction regulates expression of the anti-apoptotic molecule Bcl-xL.

Applications Tested: Tested on mouse CD254-transfected cells; use ≤0.25 µg per test (100 µL final volume). Optimize antibody titration for best performance.


Target Information

This gene encodes a member of the TNF cytokine family, a ligand for osteoprotegerin and a key factor in osteoclast differentiation and activation. It functions as a dendritic cell survival factor and regulates T cell–dependent immune responses.
T cell activation induces expression of this gene, promoting osteoclastogenesis and bone loss. It activates anti-apoptotic kinase AKT/PKB via SRC and TRAF6 signaling.
Gene disruption in mice causes osteopetrosis, lack of osteoclasts, and defects in T/B lymphocyte differentiation and mammary gland development. Two alternatively spliced transcript variants are known.
