Target Information

Methylation of DNA at cytosine residues in mammalian cells is a heritable, epigenetic modification critical for proper regulation of gene expression, genomic imprinting, and development. Three families of mammalian DNA methyltransferases have been identified: DNMT1, DNMT2, and DNMT3.
DNMT1 acts as a maintenance methyltransferase, while DNMT3A and DNMT3B function as de novo methyltransferases, strongly expressed in embryonic stem cells. DNMT2 shows low expression in adult tissues.
DNMT3L is a catalytically inactive regulatory factor for DNMT3A and DNMT3B, expressed at low levels in embryonic stem cells, testis, ovaries, and thymus. It forms a heterotetrameric complex with DNMT3A or DNMT3B and contains an N-terminal ADD domain and a C-terminal methyltransferase-like domain.
