Target Information

Translocation of nuclear-encoded preproteins into the mitochondrial matrix requires coordinated action of the translocases Tom and Tim, located in the outer and inner mitochondrial membranes, respectively.
The Tom complex includes at least eight proteins: import receptors (Tom70, Tom37, Tom22, Tom20), small proteins (Tom7, Tom6, Tom5), and the structural channel component (Tom40).
The Tim channel imports nuclear-encoded mitochondrial preproteins and involves Tim17, Tim23, and Tim44 in equimolar ratios. Tim17 exists as two isoforms (Tim17a, Tim17b) differing in their C-terminal sequences and, like Tim23, is ubiquitously expressed in fetal and adult tissues.
Tim17 and Tim23 form integral membrane proteins composing the structural elements of the inner membrane channel. Tim44 is a hydrophilic protein that recruits mitochondrial Hsp70 at the site where preproteins emerge from the Tim channel.
