Target Information
This gene generates several transcript variants differing in their first exons. At least three alternatively spliced variants encoding distinct proteins have been reported, two of which encode isoforms functioning as inhibitors of CDK4 kinase. Another transcript includes an alternate first exon located 20 Kb upstream, encoding the ARF protein that stabilizes p53 by sequestering the E3 ubiquitin-protein ligase MDM2. Despite structural and functional differences, both CDK inhibitor isoforms and the ARF product share a role in cell cycle G1 control. This gene is frequently mutated or deleted in various tumors and is an important tumor suppressor gene.
