Target Information

This gene generates several transcript variants differing in their first exons. At least three alternatively spliced variants encoding distinct proteins have been reported, two of which encode structurally related isoforms functioning as inhibitors of CDK4 kinase.
Another transcript includes an alternate first exon located 20 Kb upstream, containing an alternate open reading frame (ARF) that encodes a structurally unrelated protein. This ARF product stabilizes the tumor suppressor protein p53 by interacting with and sequestering the E3 ubiquitin-protein ligase MDM2, preventing p53 degradation.
Despite structural and functional differences, the CDK inhibitor isoforms and the ARF product share a common role in regulating cell cycle G1 progression via CDK4 and p53 pathways.
Mutations or deletions in this gene are frequent in various tumors, highlighting its importance as a tumor suppressor gene.
