Target Information
The CDKN2A gene produces several transcript variants differing in their first exons. At least three alternatively spliced variants encode distinct proteins, two of which act as inhibitors of CDK4 kinase. Another variant encodes an alternate open reading frame (ARF) protein that stabilizes tumor suppressor p53 by sequestering MDM2, preventing p53 degradation. Despite structural differences, both CDK inhibitor isoforms and the ARF product contribute to cell cycle G1 control through regulation of CDK4 and p53. CDKN2A is frequently mutated or deleted in various tumors, serving as an important tumor suppressor gene.
