Target Information
Glycogen Synthase Kinase-3 (GSK3) is a protein kinase originally identified as a regulator of glycogen synthase, a key enzyme in glycogen metabolism.
It regulates various cellular processes including:
- Protein synthesis
- Cell proliferation and differentiation
- Microtubule assembly/disassembly
- Apoptosis
GSK3 phosphorylates substrates only when a phosphoserine or phosphotyrosine residue is present four residues downstream of the target site.
There are two isoforms, GSK3 alpha and GSK3 beta, regulated via phosphorylation:
- Inactivation occurs by phosphorylation at Ser9 (Ser21 in GSK3α) by PKB.
- Activation occurs via phosphorylation at Tyr216 (Tyr279 in GSK3α).
Activated GSK3β phosphorylates multiple cellular proteins including p53, c-Myc, c-Jun, HSF-1, and cyclin D1, and is implicated in tau phosphorylation relevant to Alzheimer’s disease progression.
