Circadian rhythmicity is a fundamental property of diverse organisms, including animals, plants, and fungi. Regulation of endogenous biological clocks occurs at the genetic level via protein-mediated autoregulatory feedback loops.
In mammals, several genes encoding members of the bHLH-PAS (PER-ARNT-SIM) transcription factor family play key roles in circadian oscillations. Transactivation of CLOCK-induced genes is mediated through an E-box enhancer (CACGTG) located upstream of target genes.
CLOCK-ARNT3 heterodimers bind to E-box elements and stimulate gene transcription. CLOCK has been shown to transactivate the mammalian homolog of Drosophila per. PER, together with the product of the mammalian timeless gene (TIM), negatively regulates its own transcription by blocking the CLOCK-BMAL1 transactivation complex.
