Product Specific Information
The eBioSN5c monoclonal antibody recognizes human CD10 (CALLA, NEP, enkephalinase, Neprilysin), a 100 kDa type II cell surface glycoprotein originally identified for its expression on most acute lymphoblastic leukemias (ALLs). CD10 is identical to neutral endopeptidase (NEP) and acts as a Zn²⁺-dependent metallopeptidase acting on substrates such as endothelin, glucagon, gastrin, neurotensin, and bradykinin. It is involved in regulating chemotactic and inflammatory processes involving neutrophils and plays a role in stromal cell-dependent B lymphopoiesis. Expression is found on mature germinal center B cells, granulocytes, and bone marrow stromal cells. CB-CALLA and eBioSN5c likely recognize different epitopes due to co-staining ability.
Applications Reported: Suitable for flow cytometric analysis and immunohistology staining of frozen tissue sections.
Applications Tested: Validated for flow cytometric analysis at ≤0.25 µg/test (defined as staining a cell sample in 100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test. Antibody titration recommended for optimal assay performance.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
CD10 (Common Acute Lymphocytic Leukemia Antigen, CALLA) is a cell surface enzyme with neutral metalloendopeptidase activity that inactivates various biologically active peptides. It is expressed on lymphoblastic, Burkitt’s, and follicular germinal center lymphomas, immature B cells in bone marrow, and cells from patients with chronic myelocytic leukemia (CML). CD10 is also found on breast myoepithelial cells, bile canaliculi, fibroblasts, and highly expressed on kidney and gut epithelial brush borders. It cleaves peptides at the amino side of hydrophobic residues and inactivates peptide hormones including glucagon, enkephalins, substance P, neurotensin, oxytocin, and bradykinin. CD10 is a 100 kDa type II transmembrane glycoprotein encoded by a >45 kb gene with alternatively spliced 5′ untranslated regions resulting in four mRNA transcripts. Diseases associated with CD10 dysfunction include spinocerebellar ataxia 43 and Charcot-Marie-Tooth disease.
