This Bcl-2/100 monoclonal antibody reacts to human Bcl-2 (B-cell Lymphoma 2), the founding member of a family of pro-apoptotic proteins including Bcl-XL, Bcl-w, and Mcl-1. Bcl-2 protects against apoptosis-inducing insults such as dexamethasone, staurosporine, irradiation, and cytokine withdrawal.
Its mechanism involves regulation of mitochondrial membrane permeability, preventing release of apoptosis-mediating factors like cytochrome c and AIF into the cytoplasm.
Bcl-2 localizes to the cytoplasmic side of the mitochondrial membrane, nuclear envelope, and endoplasmic reticulum.
Two isoforms (α and β) are identified by alternative splicing at the C-terminus, and this antibody recognizes both.

Target Information

BCL-2 is a key regulator of apoptosis, functioning to inhibit or promote cell death.
It forms dimers with other family members to modulate apoptosis. For example, Bag1 forms a heterodimer with BCL-2, enhancing its anti-apoptotic effect.
Bax and Bak promote cytochrome c release from mitochondria, initiating apoptosis.
Constitutive expression of BCL-2, such as translocation to the Ig heavy chain locus, is associated with follicular lymphoma.
In most follicular lymphomas, neoplastic germinal centers express high levels of BCL-2 alpha protein, unlike normal centers.
Two transcript variants exist, differing at the C-terminal ends. Overexpression of BCL-2 is linked to cancers such as B-cell lymphoma and prostate cancer.
